Overview

siRNA-il10rb is a small interfering RNA (siRNA) designed to silence the expression of the Interleukin 10 receptor subunit beta (IL10RB). Research presented at AACR 2025 by the National Cancer Center Research Institute (Japan) identifies IL10RB as a critical regulator of tumor escape following Snail-induced epithelial-mesenchymal transition (EMT). In preclinical melanoma models, treatment with siRNA-il10rb significantly reduced cancer cell invasion, adhesion, and chemoresistance. The study suggests that IL10RB promotes the formation of invadopodia and contributes to immune dysfunction by reducing T cell and NK cell activity in the tumor microenvironment. Targeting IL10RB via siRNA or blocking antibodies is proposed as a strategy to overcome tumor intractability post-EMT.
