**SLATE-KRAS** is an investigational, off-the-shelf therapeutic cancer vaccine developed from Gritstone bio's shared neoantigen vaccine platform. It is an optimized heterologous prime-boost regimen that uses a chimpanzee adenovirus 68 vector prime and a self-amplifying mRNA boost to encode KRAS-derived tumor neoantigens. The vaccine is intended to stimulate KRAS mutation-specific antitumor T-cell responses in patients whose tumors and HLA type match the encoded neoantigens. It was developed after the earlier multivalent SLATE vaccine showed preferential immune responses to TP53 neoantigens, prompting a KRAS-focused redesign. SLATE-KRAS was evaluated in Phase 2 within NCT03953235 and is also being studied with autologous KRAS-reactive T-cell receptor-engineered peripheral blood lymphocytes in an NCI-led Phase 1 trial, NCT06253520.
