SLATEv1 is an investigational, off-the-shelf shared-neoantigen therapeutic cancer-vaccine regimen developed by Gritstone bio for advanced or metastatic solid tumors. It uses a heterologous intramuscular prime-boost approach comprising a chimpanzee adenovirus 68 vector prime followed by a self-amplifying mRNA boost, encoding 20 shared tumor neoantigens derived from recurrent mutations including those in KRAS, TP53, BRAF, and CTNNB1. The intended mechanism is induction of neoantigen-specific antitumor T-cell responses; it was clinically evaluated with nivolumab and ipilimumab, which are separate co-administered checkpoint inhibitors rather than components of SLATEv1 itself.
