SN27686 is a dinitrobenzamide bromomustard prodrug developed for use in directed enzyme prodrug therapy (DEPT) applications, such as gene-directed enzyme prodrug therapy (GDEPT) and bacterial-directed enzyme prodrug therapy (BDEPT). The compound is designed to be relatively non-toxic until it is selectively reduced by nitroreductase (NTR) enzymes, which are delivered to the tumor site via viral or bacterial vectors. Upon reduction of its nitro groups, SN27686 is converted into a highly reactive DNA-alkylating species that induces genotoxic stress and apoptosis. Research has demonstrated its efficacy in chemoresistant small cell lung cancer (SCLC) and ovarian cancer models, particularly when used in combination with other suicide gene therapy systems or magnetic nanoparticle-directed delivery.
