SOID-8 is a synthetic spirooxindole derivative and an analogue of the natural product spirotryprostatin B. It functions as a small molecule inhibitor of the JAK2/STAT3 signaling pathway. Developed by researchers at the City of Hope, SOID-8 has demonstrated preclinical activity against melanoma by inhibiting cell proliferation and inducing apoptosis in human melanoma cell lines such as A2058 and A375. Its mechanism of action involves the dose- and time-dependent inhibition of Janus kinase 2 (JAK2) and Signal Transducer and Activator of Transcription 3 (STAT3) phosphorylation, which subsequently leads to the downregulation of the anti-apoptotic protein Mcl-1 and the induction of PARP cleavage.
