Sonrotoclax + rituximab is an investigational combination regimen under clinical development for the treatment of relapsed or refractory chronic lymphocytic leukemia (CLL), small lymphocytic lymphoma (SLL), and mantle cell lymphoma (MCL). **Sonrotoclax** is a small molecule inhibitor of B-cell lymphoma-2 (BCL-2), a protein that promotes cell survival in certain hematological malignancies. By inhibiting BCL-2, sonrotoclax induces apoptosis in malignant B-cells. **Rituximab** is a monoclonal antibody targeting the CD20 antigen on B-cells, leading to B-cell depletion through multiple immune-mediated mechanisms. The combination aims to enhance anti-tumor efficacy beyond each agent alone, exploiting complementary mechanisms of BCL-2 inhibition and targeted immunotherapy. This combination is currently being studied in phase 2 and phase 3 clinical trials for B-cell malignancies, particularly relapsed/refractory CLL/SLL[1][2][3][4][5][6][9][10].
