SP-101 (formerly CP-001) is a polymer-drug conjugate consisting of the glucocorticoid dexamethasone chemically conjugated to a hydroxypropyl methacrylate (HPMA) copolymer. Originally developed by Chariton Pharmaceuticals and later acquired by Savina Pharma, the drug is designed for the targeted delivery of dexamethasone to inflamed tissues, specifically for the treatment of rheumatoid arthritis. By utilizing the enhanced permeability and retention (EPR) effect in inflamed joints, SP-101 aims to provide localized activation of glucocorticoid receptors. This targeted approach is intended to maintain the potent anti-inflammatory efficacy of dexamethasone while significantly reducing the systemic side effects typically associated with long-term corticosteroid use. Preclinical studies in rodent models of collagen-induced arthritis have demonstrated robust efficacy and a superior safety profile compared to systemic dexamethasone administration.
