Overview

SPH1025 (also known as 1025-A) is an orally bioavailable, small molecule inhibitor that selectively targets Janus kinase 1 (JAK1). Developed by Shanghai Pharmaceuticals Holding Co., Ltd., it is designed to treat autoimmune and inflammatory diseases by modulating the JAK-STAT signaling pathway. This pathway is a critical mediator for various pro-inflammatory cytokines, such as interleukin-6 (IL-6) and interferons, which drive the pathogenesis of conditions like rheumatoid arthritis. By specifically inhibiting JAK1, SPH1025 aims to provide therapeutic efficacy in reducing joint inflammation and damage while potentially offering a superior safety profile compared to non-selective JAK inhibitors. The drug is currently in Phase 2 clinical development for the treatment of arthritis.
