Stabilized anti-CD19 CAR mRNA is an in vivo mRNA-based cell therapy candidate developed by ParcelBio. It utilizes the company's proprietary "Stabilizer Elements" platform, which incorporates modular sequence elements into the mRNA to significantly enhance protein expression levels and durability, achieving circular RNA-like stability. The therapy is designed for intravenous delivery via targeted lipid nanoparticles (LNPs) that specifically reprogram T cells in vivo to express a chimeric antigen receptor (CAR) targeting CD19. This approach aims to provide a transient yet potent and durable B-cell depletion, offering a safer and more scalable alternative to traditional ex vivo integration-based CAR-T therapies. Preclinical data presented at ASGCT 2026 demonstrated that this stabilized mRNA design achieved superior CAR expression and complete B-cell clearance in humanized mouse models compared to standard clinical mRNA designs.
