This is an experimental **STING-boosted mRNA cancer nanovaccine platform** developed at King's College London. The reported formulation co-encapsulates mRNA encoding the model tumor antigen ovalbumin and the cyclic dinucleotide STING agonist 2′3′-cGAMP in an ionizable stable nucleic acid lipid nanoparticle. The mRNA component promotes antigen expression and presentation by dendritic cells, while 2′3′-cGAMP activates STING-mediated innate immune signaling, enhancing dendritic-cell maturation and antigen-specific CD8-positive T-cell responses. In B16F10-OVA murine melanoma models, the formulation showed prophylactic tumor prevention and therapeutic tumor-control activity following intratumoral or intramuscular administration.
