The surrogate anti-mouse IL1RAP ADC is a research-grade antibody-drug conjugate (ADC) developed by Cantargia AB in collaboration with AbbVie. It consists of a monoclonal antibody specific for murine Interleukin-1 receptor accessory protein (IL1RAP) conjugated to a maytansinoid cytotoxic payload via a cleavable linker. This surrogate molecule was designed to evaluate the safety and toxicity profile of IL1RAP-targeting ADCs in immunocompetent mouse models, as the primary clinical candidate is optimized for human and cynomolgus IL1RAP. IL1RAP is a co-receptor for IL-1, IL-33, and IL-36 signaling and is overexpressed on various tumor cells and within the tumor microenvironment, including cancer-associated fibroblasts and suppressive immune cells. The surrogate allows for the assessment of potential on-target, off-tumor toxicities in tissues where IL1RAP is endogenously expressed in mice.
