Overview

SW034510 is a small molecule compound identified by the NCI Cancer Treatment Discovery and Development Network (CTD2N) consortium, involving researchers from the University of Texas Southwestern Medical Center and other institutions. It was discovered through a screen of chemical libraries for agents that selectively kill lung cancer cells while sparing normal lung epithelial cells. SW034510 demonstrates potent growth inhibition in both small cell lung cancer (SCLC) and non-small cell lung cancer (NSCLC) models. Mechanistically, the compound induces apoptosis, as evidenced by the presence of cleaved caspase-3. Notably, its efficacy profile in SCLC cell lines does not correlate with the response to standard-of-care agents such as etoposide or topotecan, and it appears effective across various molecular subtypes of SCLC, including those defined by ASCL1, NeuroD1, and Myc family expression. This suggests that SW034510 may offer a novel therapeutic approach for patients with recalcitrant or treatment-resistant lung cancers.
