T cell-LNP Gene Writer (CD20 CAR) is an experimental in vivo CAR-T therapy being developed by Tessera Therapeutics. The drug utilizes a targeted lipid nanoparticle (LNP) delivery system to transport an all-RNA payload—comprising Gene Writer mRNA and a CD20-specific chimeric antigen receptor (CAR) template—directly to T cells within the body. Once delivered, the Gene Writer protein uses target-primed reverse transcription to stably integrate the CAR transgene into the T cell genome. This approach aims to generate functional CAR-T cells in vivo, bypassing the complexities of ex vivo manufacturing. Preclinical studies in humanized mouse models have demonstrated significant CAR-T cell expansion and systemic B-cell depletion, indicating potential efficacy for B-cell malignancies and autoimmune diseases.
