Drug intelligence / Profile preview
Monjuvi + lenalidomide + rituximab + cyclophosphamide + doxorubicin + vincristine + prednisone (Tafa-Len-R-CHOP) is an investigational seven-drug combination regimen being evaluated as a first-line treatment for patients with newly diagnosed, high-risk diffuse large B-cell lymphoma (DLBCL) or high-grade B-cell lymphoma (HGBL). The regimen combines the standard-of-care R-CHOP immunochemotherapy backbone (rituximab, cyclophosphamide, doxorubicin, vincristine, and prednisone) with tafasitamab (Monjuvi), an Fc-modified cytolytic CD19-targeting monoclonal antibody, and lenalidomide, an immunomodulatory drug. Tafasitamab and lenalidomide act synergistically to enhance antibody-dependent cellular cytotoxicity (ADCC) and phagocytosis (ADCP) against malignant B cells. In the pivotal Phase III frontMIND trial, the addition of tafasitamab and lenalidomide to R-CHOP significantly improved progression-free survival (PFS) compared to R-CHOP alone, reducing the risk of disease progression or death by 25%.
