TAT-RNF187 is a cell-permeable recombinant fusion protein being investigated for the treatment of inflammatory bowel disease (IBD). It consists of the RING finger 187 (RNF187) protein, also known as RING domain AP-1 co-activator-1, fused to a TAT (trans-activator of transcription) cell-penetrating peptide to facilitate intracellular delivery. Mechanistically, TAT-RNF187 acts as a negative regulator of intestinal inflammation by targeting the IKK complex. Specifically, the C-terminal 163-265 domain of RNF187 interacts with the leucine zipper (LZ) domains of both IKKα and IKKβ, thereby antagonizing the formation and subsequent activation of the IKK complex. This inhibition suppresses the NF-κB signaling pathway, which is a central driver of inflammation in IBD. Preclinical studies in mouse models of colitis have demonstrated that TAT-RNF187 can alleviate intestinal inflammation and may enhance the therapeutic efficacy of biological agents such as infliximab.
