Overview

This is an investigational combination therapy comprised of **tazemetostat**, an orally available small molecule inhibitor of the enzyme EZH2 (Enhancer of Zeste Homolog 2), and **mosunetuzumab**, a bispecific monoclonal antibody that targets CD20 on B cells and CD3 on T cells. Tazemetostat works by inhibiting EZH2, a methyltransferase involved in epigenetic gene silencing, which is implicated in the proliferation and survival of B-cell lymphomas. Inhibition of EZH2 slows lymphoma growth and may enhance anti-tumor immune responses. Mosunetuzumab is designed to redirect T-cells to recognize and destroy CD20-positive lymphoma cells by bridging CD20 and CD3, functioning as an immunotherapy. The combination is being studied for the treatment of untreated follicular lymphoma to explore potential additive or synergistic efficacy compared to either agent alone. Tazemetostat is FDA-approved for use in relapsed/refractory follicular lymphoma, and mosunetuzumab is approved for intravenous use in relapsed/refractory follicular lymphoma, but the combination is investigational and not yet FDA-approved[1][3][5].
