TCE-A is an investigational, Fc-silenced, IgG1-based trispecific T-cell engager (TCE) designed to simultaneously target CD19 and CD20 on B-cells and CD3 on T-cells. Developed to address limitations of single-target therapies such as antigen loss and tumor heterogeneity, TCE-A utilizes an "OR-gate" mechanism, allowing for potent B-cell depletion even if one target is downregulated. The molecule incorporates an affinity-tuned CD3 binder to potentially reduce cytokine release syndrome (CRS) and a unique CD20 binder with an epitope distinct from rituximab, making it compatible with rituximab-based combination therapies. Preclinical data demonstrate high efficacy in B-cell non-Hodgkin lymphoma (B-NHL) models and systemic lupus erythematosus (SLE) models, showing significant reduction in pathogenic B-cells and autoantibodies. Its pharmacokinetic profile supports subcutaneous administration with a half-life of approximately 4-5 days.
