TCR-T cells + IL-15(D8S) is an experimental cell therapy being developed by Regeneron Pharmaceuticals. It consists of T cells engineered to express a specific T cell receptor (TCR) alongside a modified version of interleukin-15 (IL-15) containing a D8S mutation. This mutation reduces the cytokine's affinity for the IL-2Rβ receptor, thereby restricting its activity to cells expressing the high-affinity IL-15Rα receptor, such as the engineered T cells themselves. To further enhance safety and prevent systemic toxicity, the expression of IL-15(D8S) is regulated by an activation-induced synthetic promoter (iSP), which triggers cytokine production only upon TCR engagement with the target tumor antigen. Preclinical data indicate that this dual-control strategy enhances T cell potency, expansion, and tumor control in xenograft models while avoiding the continuous T cell expansion seen with wild-type IL-15.
