**TD-1473 + itraconazole** is a combination of two drugs: TD-1473 (also known as izencitinib), an orally administered, gut-selective, potent pan-Janus kinase (JAK) inhibitor designed for the treatment of inflammatory bowel diseases such as ulcerative colitis, and itraconazole, a triazole antifungal medication. TD-1473 acts locally in the gastrointestinal tract to inhibit JAK-dependent proinflammatory cytokine signaling (JAK1, JAK2, JAK3, TYK2) with reduced systemic exposure, aiming to limit adverse effects associated with systemic JAK inhibition. Itraconazole inhibits fungal cell membrane synthesis via inhibition of lanosterol 14\u03b1-demethylase. The combination may be of interest for drug-drug interaction or clinical pharmacology studies, as itraconazole is a strong CYP3A4 inhibitor, often used to study the metabolic drug interactions of orally administered compounds[1][2][10].
