Overview

TfNP181a is a transferrin-conjugated nanoparticle delivery system encapsulating synthetic precursor microRNA-181a (miR-181a). Developed by researchers at The Ohio State University, it is designed to target acute myeloid leukemia (AML) cells, which often overexpress the transferrin receptor. The nanoparticle consists of a polyethylenimine (PEI) core complexed with miR-181a, surrounded by a lipid layer (DOPE, linoleic acid, and DMPE-mPEG) for protection and cellular uptake. Once delivered, miR-181a downregulates the expression of KRAS and NRAS, thereby inhibiting MAPK signaling. This mechanism sensitizes AML cells to chemotherapy agents like daunorubicin.
