TG02 refers to two distinct therapeutic agents identified in clinical oncology: a peptide-based cancer vaccine and a small-molecule multi-kinase inhibitor. The **TG02 vaccine** (developed by Targovax, now Circio) is a second-generation neoantigen immunotherapy designed to target the eight most common RAS mutations (codon 12 and 13, exon 2) found in colorectal cancer and other solid tumors. It consists of eight synthetic peptides and is typically administered with a GM-CSF adjuvant to stimulate a T-cell mediated immune response against KRAS-mutant cells. The **TG02 small molecule** (originally SB1317, developed by S*BIO and licensed to Tragara Pharmaceuticals) is an orally bioavailable multi-kinase inhibitor. It potently inhibits cyclin-dependent kinases (primarily CDK9, but also CDK1, CDK2, CDK5, and CDK7), as well as JAK2 and FLT3. This small molecule is studied for its ability to disrupt transcriptional programs and deplete short-lived survival proteins like Mcl-1, showing potential in treating glioblastoma and hematologic malignancies.
