TG21 is a therapeutic cancer vaccine candidate designed to target mutant RAS proteins, which are prevalent drivers in various solid tumors. Developed as part of a second-generation platform following TG01, TG21 consists of a mixture of synthetic peptides representing common RAS mutations (such as G12D, G12V, and G12R) combined with an adjuvant to stimulate a robust T-cell response. The mechanism of action involves the induction of mutation-specific cytotoxic T lymphocytes (CTLs) and helper T cells that recognize and eliminate cancer cells expressing the targeted neoantigens. TG21 is primarily investigated for the treatment of KRAS-mutated cancers, including pancreatic, colorectal, and lung adenocarcinomas, often in combination with other immunotherapies or standard-of-care chemotherapy to enhance clinical efficacy.
