TGW211-DC

**TGW211-DC** is a HER2-directed, click-cleavable radioimmunoconjugate developed by Tagworks using trastuzumab functionalized with a DOTA chelator through a trans-cyclooctene linker. After radiolabeling, the conjugate binds and internalizes into HER2-positive tumor cells. A subsequently administered non-cell-permeable tetrazine trigger reacts with residual circulating conjugate, releasing a small radiolabeled DOTA-containing fragment for rapid renal clearance while retaining radioactivity within tumor cells. The TGW211 program is being evaluated in the Phase 0/1 CleavHER study in HER2-positive solid tumors, initially using indium-111 imaging and with preclinical therapeutic applications using lutetium-177 and actinium-225.
