TJ111 is a preclinical-stage bispecific, dual-payload antibody-drug conjugate (ADC) being developed by Phrontline Biopharma for the treatment of urothelial cancer. The molecule is engineered to simultaneously target two cell surface antigens, Tumor-associated calcium signal transducer 2 (Trop2) and Nectin cell adhesion molecule 4 (Nectin4), which are highly co-expressed in urothelial malignancies. TJ111 utilizes a novel dual-payload platform to deliver both a topoisomerase I inhibitor and a microtubule inhibitor directly to the tumor site. This dual-targeting and dual-cytotoxic approach aims to enhance therapeutic precision, increase payload delivery efficiency, and potentially mitigate the development of drug resistance commonly observed with single-target or single-payload therapies.
