Overview

A TNFR2 neutralizing antibody is a type of monoclonal antibody designed to bind to Tumor Necrosis Factor Receptor 2 (TNFR2, also known as TNFRSF1B or CD120b) and block its activation by TNFα. Unlike TNFR1, which is ubiquitously expressed, TNFR2 expression is largely restricted to certain immune cell subsets, such as regulatory T cells (Tregs) and myeloid-derived suppressor cells (MDSCs), as well as various tumor cells. By neutralizing TNFR2 signaling, these antibodies aim to deplete or inhibit the suppressive function of Tregs within the tumor microenvironment and directly inhibit the growth of TNFR2-expressing cancer cells. This approach is being investigated as a potent cancer immunotherapy, often in combination with PD-1/PD-L1 inhibitors, to overcome immune resistance.
