Overview

This treatment group represents a combination of immunomodulatory therapies evaluated in an observational study by Aetion, Inc. for the treatment of hospitalized COVID-19 patients. The group includes two classes of agents: interleukin-6 (IL-6) receptor inhibitors (tocilizumab and sarilumab) and Janus kinase (JAK) inhibitors (baricitinib and tofacitinib), all administered in conjunction with systemic corticosteroids (e.g., dexamethasone). Tocilizumab and sarilumab are monoclonal antibodies that bind to the IL-6 receptor, preventing the pro-inflammatory effects of IL-6. Baricitinib and tofacitinib are small-molecule inhibitors that target the JAK-STAT signaling pathway, which is central to the cytokine storm observed in severe COVID-19. Systemic corticosteroids provide broad anti-inflammatory effects. This specific combination/comparison is used to determine the relative effectiveness of IL-6 inhibitors versus JAK inhibitors in reducing mortality and the need for mechanical ventilation in patients already receiving steroid therapy.
