Baricitinib and tofacitinib are small-molecule inhibitors that target the JAK-STAT signaling pathway, which is central to the cytokine storm observed in severe COVID-19. Systemic corticosteroids provide broad anti-inflammatory effects. This specific combination/comparison is used to determine the relative effectiveness of IL-6 inhibitors versus JAK inhibitors in reducing mortality and the need for mechanical ventilation in patients already receiving steroid therapy.
