Drug intelligence / Profile preview
Tositumomab and iodine I 131 tositumomab (Bexxar) is a radioimmunotherapy regimen consisting of a murine IgG2a monoclonal antibody (tositumomab) and its iodine-131 radiolabeled counterpart. It targets the CD20 antigen expressed on the surface of B-lymphocytes. The mechanism of action involves a dual approach: the antibody-dependent cell-mediated cytotoxicity (ADCC), complement-dependent cytotoxicity (CDC), and apoptosis induction by the tositumomab antibody, combined with targeted delivery of ionizing radiation from the iodine-131 isotope, which induces DNA damage in both targeted and neighboring 'bystander' cells. Developed by GSK, it was evaluated by the SWOG Cancer Research Network in the Phase II S0433 trial as a consolidation therapy following R-CHOP chemotherapy for patients with previously untreated advanced-stage diffuse large B-cell lymphoma (DLBCL). Commercial production and availability in the United States were discontinued by GSK in 2014.
