Overview

Tr-ACT2 is a first-in-class antibody encapsulated drug (AED) developed by Sunstate Biosciences using its proprietary single protein encapsulation (SPE) platform. It consists of the HER2-targeted monoclonal antibody trastuzumab (Tr) non-covalently encapsulating two molecules of the cytotoxic payload actinomycin D (ACT), an RNA polymerase inhibitor. Unlike traditional antibody-drug conjugates (ADCs), Tr-ACT2 does not utilize chemical linkers or artificial nanoparticles, aiming to overcome challenges such as in vivo instability and suboptimal payload release. Tr-ACT2 has demonstrated potent anticancer activity in both HER2-positive and HER2-negative cancer models, including breast cancer and non-small cell lung cancer (NSCLC), by leveraging HER2-dependent internalization in early stages and HER2-independent mechanisms in later stages.
