Overview

TR422 is a topical gel formulation of the small molecule vardenafil, currently under development by Tritech Biopharmaceuticals for the treatment of pulmonary arterial hypertension (PAH). The drug acts as a phosphodiesterase-5 (PDE5) inhibitor, preventing the degradation of cyclic guanosine monophosphate (cGMP) to promote vasodilation in the pulmonary vasculature and reduce pulmonary arterial pressure. TR422 utilizes Tritech's proprietary transdermal enhancer technology, which allows the active ingredient to penetrate the skin and be stored in tissue for sustained release into the systemic circulation. This non-invasive delivery approach is designed to provide long-lasting therapeutic effects while minimizing the gastrointestinal and systemic side effects often associated with oral PDE5 inhibitors. The product is currently in Phase 1 clinical development.
