Overview

Trametinib + ruxolitinib is an investigational combination therapy being evaluated for the treatment of advanced RAS-mutant colorectal cancer and pancreatic adenocarcinoma. This regimen combines trametinib, a reversible, highly selective, allosteric inhibitor of MEK1 and MEK2 (components of the MAPK/ERK pathway), with ruxolitinib, a potent inhibitor of Janus kinases JAK1 and JAK2 (components of the JAK/STAT pathway). The National Cancer Centre, Singapore, is sponsoring a Phase Ib clinical trial (NCT04303403) to assess the safety and efficacy of this dual-pathway inhibition strategy. The combination aims to overcome the therapeutic challenges of RAS-mutated malignancies, where compensatory signaling through the STAT3 pathway often limits the effectiveness of MEK inhibitors alone.
