**Trametinib + ruxolitinib + retifanlimab** is a three-drug combination investigated for the treatment of metastatic pancreatic ductal adenocarcinoma. - **Trametinib** is a selective inhibitor of MEK1 and MEK2, integral components of the MAPK/ERK kinase pathway, blocking cell proliferation signals. - **Ruxolitinib** inhibits Janus kinase 1 (JAK1) and Janus kinase 2 (JAK2), thereby suppressing the JAK/STAT signaling pathway involved in tumor cell survival and immune evasion. - **Retifanlimab** is a monoclonal antibody that blocks programmed cell death protein 1 (PD-1), thereby restoring T-cell mediated anti-tumor immune responses. Their combined use is designed to provide triple blockade of tumor growth and immune evasion mechanisms: targeted kinase inhibition and immune checkpoint inhibition. This combination is currently being investigated in phase 1 clinical trials[1][2][3][8].
