Overview

TRICAR-ALL is an investigational trivalent chimeric antigen receptor (CAR) T-cell therapy designed to treat B-cell malignancies, specifically Acute Lymphoblastic Leukemia (ALL). The therapy involves genetically modifying a patient's own T cells using a retroviral vector to express a chimeric receptor that simultaneously targets three distinct B-cell antigens: CD19, CD20, and CD22. This multi-antigen targeting strategy is intended to prevent tumor immune escape caused by antigen loss or downregulation, a common cause of relapse in monovalent CAR-T therapies. The CAR construct also incorporates the 4-1BB (CD137) costimulatory domain, which enhances T-cell proliferation, metabolic fitness, and long-term persistence within the patient's body.
