Overview

TriCAR19.20.22 T cells is an investigational trivalent chimeric antigen receptor (CAR) T-cell therapy developed by City of Hope for the treatment of various B-cell malignancies. This therapy involves genetically engineering a patient's own T cells to express a chimeric antigen receptor that simultaneously targets three distinct surface antigens found on B cells: CD19, CD20, and CD22. This multi-antigen targeting strategy is specifically designed to overcome the challenge of antigen escape, a common mechanism of resistance and relapse observed with single-target (monovalent) CAR T-cell therapies. By broadening the range of targets, the therapy aims to achieve more durable responses in patients with relapsed or refractory B-cell cancers. It is currently being evaluated in clinical trials for patients with non-Hodgkin lymphoma (NHL), acute lymphoblastic leukemia (ALL), chronic lymphocytic leukemia (CLL), and lymphoid blast crisis from chronic myeloid leukemia (CML).
