TRU-015 is a proprietary antibody-based single-chain polypeptide therapeutic designed to target and deplete B cells by binding specifically to the CD20 antigen on their surface. It is classified as a small modular immunopharmaceutical (SMIP), which is significantly smaller than conventional monoclonal antibodies but retains both Fv fragment-type target binding and Fc fragment-type effector functions. TRU-015 induces B cell depletion through mechanisms including antibody-dependent cell-mediated cytotoxicity (ADCC), complement-dependent cytotoxicity (CDC, attenuated compared to rituximab), and apoptosis. The drug was developed for intravenous administration in autoimmune diseases such as rheumatoid arthritis and for B-cell malignancies, showing efficacy in preclinical tumor models and clinical trials for rheumatoid arthritis. TRU-015 was originally developed by Trubion Pharmaceuticals, later partnered with Wyeth Pharmaceuticals, and subsequently involved Pfizer after corporate acquisitions[1][3][5][6][8].
