TXP1-CD20mAb is a blood-brain barrier (BBB)-crossing therapeutic candidate developed by Ossianix for the treatment of multiple sclerosis. It consists of an anti-CD20 monoclonal antibody conjugated to the TXP1 VNAR, a shark-derived single-domain antibody that acts as a molecular shuttle. The TXP1 VNAR targets the transferrin receptor 1 (TfR1) to facilitate receptor-mediated transcytosis across the BBB, allowing the anti-CD20 antibody to enter the central nervous system (CNS). By delivering B-cell depleting therapy directly to the CNS, TXP1-CD20mAb aims to address the compartmentalized inflammation and B-cell follicles associated with progressive forms of multiple sclerosis more effectively than standard systemic anti-CD20 therapies, which have limited CNS penetration.
