Overview

Ublituximab is a low-fucose, CD20-directed cytolytic monoclonal antibody used for the treatment of relapsing forms of multiple sclerosis (MS), including clinically isolated syndrome, relapsing-remitting disease, and active secondary progressive disease in adults. It works by binding to the CD20 antigen on pre-B and mature B lymphocytes, leading to their depletion through mechanisms such as antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity. Ublituximab is uniquely engineered with reduced fucose content to enhance its interaction with FcγRIIIa receptors on natural killer cells and macrophages, thereby increasing ADCC activity compared to other anti-CD20 antibodies. The drug was developed by TG Therapeutics and is marketed under the brand name Briumvi[2][3][5][6].
