ublituximab + umbralisib

Ublituximab + umbralisib is a fixed combination therapy used in the treatment of B-cell malignancies, particularly chronic lymphocytic leukemia (CLL) and non-Hodgkin lymphoma (NHL). Ublituximab is a next-generation, glycoengineered type 1 chimeric monoclonal antibody that targets a unique epitope on the B‑lymphocyte antigen CD20 found on B cells. This engineering enhances antibody-dependent cellular cytotoxicity compared to earlier anti-CD20 antibodies. Umbralisib is an oral small molecule inhibitor targeting phosphatidylinositol 4,5-bisphosphate 3‑kinase catalytic subunit delta isoform (PI3Kδ) and casein kinase I isoform epsilon (CK1ε), both key regulators in B-cell receptor signaling pathways. The combination has demonstrated synergistic activity in preclinical studies and showed promising efficacy and safety profiles in clinical trials for relapsed/refractory CLL and NHL. However, due to concerns about overall survival imbalance observed in phase 3 trials, applications for approval were voluntarily withdrawn by the developer[1][3][5][7].
