Overview

A combination regimen of two novel agents—ublituximab, a glycoengineered anti-CD20 monoclonal antibody, and umbralisib, a dual phosphoinositide 3-kinase delta (PI3Kδ) and casein kinase 1 epsilon (CK1ε) inhibitor—together with TG-1501 (cosibelimab), an anti-PD-L1 monoclonal antibody, under investigation for the treatment of various B-cell malignancies and potentially autoimmune/inflammatory disease. Ublituximab targets CD20 on B-cells, inducing cytotoxicity and apoptosis. Umbralisib selectively inhibits PI3Kδ and CK1ε, key targets in B-cell malignancies and immune regulation. TG-1501 (cosibelimab) blocks PD-L1-mediated immune evasion in the tumor microenvironment. The combination is being explored for efficacy and tolerability, aiming for improved disease control and reduced toxicities compared to existing therapies[1][3][4][5][8].
