**UCART-CLL1.BiTE-Siglec6** is an investigational, off-the-shelf allogeneic CAR-T cell therapy being developed for acute myeloid leukemia. The engineered donor-derived T cells express a chimeric antigen receptor directed against C-type lectin-like molecule-1 and continuously secrete a Siglec-6 × CD3 bispecific T-cell engager. This dual-target strategy is intended to directly eliminate CLL-1-positive AML blasts while recruiting both administered CAR-T cells and endogenous T cells against Siglec-6-positive leukemia cells, potentially reducing antigen-escape risk. The reported preclinical construct uses CLL-1 and Siglec-6 nanobodies and was designed to target AML blasts and leukemia stem cells while sparing hematopoietic stem cells.
