UCART20x22 is a first-in-class, allogeneic, non-alloreactive, engineered human CAR T-cell product developed by Cellectis. It is designed with dual chimeric antigen receptor (CAR) expression to simultaneously target CD20 and CD22 antigens, aiming to overcome antigen escape in B-cell malignancies. The product is manufactured using TALEN® gene-editing technology to inactivate the *TRAC* gene, which reduces the risk of graft-versus-host disease (GvHD), and the *CD52* gene, which allows the cells to persist in the presence of alemtuzumab-based lymphodepletion regimens. UCART20x22 is currently being evaluated in the Phase 1/2a Nathali-01 clinical trial for patients with relapsed or refractory B-cell non-Hodgkin lymphoma (B-NHL), including those who have failed prior CD19-directed CAR-T therapies. Preclinical data suggests that combining UCART20x22 with low-dose interleukin-2 (IL-2) can enhance the expansion and persistence of the CAR-T cells, potentially improving therapeutic outcomes.
