Overview

ABBV-722 + upadacitinib is an investigational combination therapy being developed by AbbVie for the treatment of various inflammatory diseases. The combination consists of upadacitinib, a selective Janus kinase 1 (JAK1) inhibitor, and ABBV-722, a next-generation selective glucocorticoid receptor modulator (SGRM). Upadacitinib is an approved therapy (marketed as Rinvoq) that blocks the JAK-STAT signaling pathway, thereby reducing the production of pro-inflammatory cytokines. ABBV-722 is designed to activate the glucocorticoid receptor to induce anti-inflammatory effects while potentially minimizing the metabolic and systemic side effects typically associated with traditional high-dose corticosteroids. By targeting two distinct inflammatory pathways, the combination aims to provide enhanced clinical efficacy for immune-mediated conditions. It is currently being evaluated in Phase 1 clinical trials to assess safety and pharmacokinetic interactions.
