**VEGFC-vax** is an experimental, lymphangiogenesis-inducing whole-cell melanoma vaccine developed in a preclinical mouse model. It consists of lethally irradiated B16F10 or B16F10-OVA melanoma cells genetically engineered to overexpress vascular endothelial growth factor C, formulated with topically retained immune adjuvants. Following intradermal administration, VEGFC-vax promotes local lymphatic-vessel growth and lymphatic transport to draining lymph nodes, increases CCL21 expression and recruitment of CCR7-positive naïve T cells, and elicits broad melanoma antigen-specific T-cell immunity. In prophylactic mouse studies, it improved protection against melanoma tumor challenge relative to a control whole-cell vaccine and a GM-CSF-secreting whole-cell vaccine.
