Overview

Veltuzumab is a humanized IgG1 type I monoclonal antibody that targets the CD20 antigen expressed on B lymphocytes. It was developed as an anti-CD20 therapy with enhanced binding avidity and increased complement-dependent cytotoxicity (CDC) compared to rituximab. Veltuzumab induces cell death in CD20-expressing cells through mechanisms including complement-dependent cell lysis (CDCL) and antibody-dependent cell-mediated cytotoxicity (ADCC). It differs from rituximab by only one amino acid in the complementarity-determining region but has a distinct framework that results in lower off-rates and improved CDC activity. The drug was primarily investigated for non-Hodgkin's lymphoma, chronic lymphocytic leukemia (CLL), idiopathic thrombocytopenic purpura (ITP), pemphigus, and rheumatoid arthritis[1][2][3][4][5][6][7].
