Veltuzumab-SN-38 (also known by the code name IMMU-110) is an antibody-drug conjugate (ADC) developed by **Immunomedics** (subsequently acquired by **Gilead Sciences**) for the treatment of various B-cell malignancies, including non-Hodgkin lymphoma and leukemia. The drug consists of veltuzumab, a humanized second-generation anti-CD20 monoclonal antibody, conjugated to the cytotoxic payload SN-38, which is the active metabolite of the chemotherapy drug irinotecan. The ADC utilizes the proprietary pH-sensitive CL2A linker, which is designed to allow the controlled release of SN-38 both within the tumor cell after internalization and in the tumor microenvironment (bystander effect). By targeting CD20, the conjugate delivers the potent topoisomerase I inhibitor directly to malignant B-cells, leading to DNA damage and apoptosis. While it showed promising preclinical and early clinical activity, its development was not prioritized following the acquisition of Immunomedics.
