Venetoclax + ibrutinib + rituximab is a combination therapy regimen being investigated for the treatment of relapsed or refractory diffuse large B-cell lymphoma (DLBCL). The regimen combines venetoclax, a selective BCL-2 inhibitor that restores the apoptotic process in cancer cells; ibrutinib, a covalent BTK inhibitor that blocks B-cell receptor signaling pathways essential for malignant B-cell survival; and rituximab, an anti-CD20 monoclonal antibody that facilitates immune-mediated clearance of B-cells. This triplet approach, spearheaded by Hackensack Meridian Health in clinical trials, aims to overcome resistance mechanisms and improve clinical outcomes in aggressive non-Hodgkin lymphomas by simultaneously targeting multiple survival nodes.
