venetoclax + rituximab

Venetoclax + rituximab is a fixed-duration combination therapy used primarily for the treatment of adults with chronic lymphocytic leukemia (CLL) or small lymphocytic lymphoma (SLL), including relapsed or refractory cases. Venetoclax is a first-in-class small molecule inhibitor of B-cell lymphoma 2 (BCL-2), an anti-apoptotic protein overexpressed in CLL cells; by inhibiting BCL-2, venetoclax restores apoptosis in malignant B-cells. Rituximab is a monoclonal antibody targeting CD20 on the surface of B-cells and mediates cell death through multiple mechanisms including complement-dependent cytotoxicity and antibody-dependent cellular cytotoxicity. The combination has demonstrated superior efficacy compared to standard chemoimmunotherapy regimens in clinical trials such as MURANO[1][3][5]. Venetoclax was developed by AbbVie and Genentech/Roche; rituximab was developed by Biogen and Genentech/Roche.
