Voyager-V1 + ruxolitinib is an investigational combination therapy consisting of Voyager-V1 (VSV-hIFNbeta-NIS), an engineered oncolytic vesicular stomatitis virus (VSV), and ruxolitinib, a small molecule JAK1/JAK2 inhibitor. Developed by Vyriad in partnership with Mayo Clinic, Voyager-V1 is designed to selectively replicate in and destroy cancer cells while expressing human interferon-beta (IFNβ) to stimulate antitumor immunity and the sodium iodide symporter (NIS) to enable SPECT/PET-based tracking of viral spread. Ruxolitinib is co-administered to transiently inhibit the JAK/STAT pathway, thereby dampening innate antiviral interferon signaling and enhancing the replication, spread, and oncolytic efficacy of Voyager-V1 within the tumor microenvironment. The combination is being evaluated in Phase 1 clinical trials for the treatment of relapsed/refractory multiple myeloma, acute myeloid leukemia, T-cell lymphoma, and stage IV or recurrent endometrial cancer.
