VX-11e is a potent, selective, and ATP-competitive small molecule inhibitor of extracellular signal-regulated kinases 1 and 2 (ERK1/2). Developed by Vertex Pharmaceuticals, it is a well-characterized pharmacological tool used to study the MAPK/ERK signaling pathway, which regulates critical cellular processes such as proliferation, differentiation, and survival. VX-11e is frequently employed in preclinical research as a positive control for ERK inhibition. It has been utilized in studies investigating airway smooth muscle hyperplasia in asthma and the microenvironment of chondrocytes and osteoclasts in osteoarthritis and osteoporosis. In clinical research, it has been featured in exploratory Phase 0 studies as a reference compound for evaluating the effects of other targeted therapies on patient-derived tissue samples.
