Overview

WP1066 is an orally bioavailable, small molecule inhibitor of signal transducer and activator of transcription 3 (STAT3), developed as a potential antineoplastic and immunomodulatory agent. It blocks the intranuclear translocation of phosphorylated STAT3, suppressing STAT3 signaling and reducing downstream oncogenic products such as c-myc. This inhibition can decrease tumor cell proliferation, angiogenesis, metastasis, and immune evasion. Additionally, WP1066 may upregulate costimulatory molecules on microglia and reverse cancer stem cell-mediated immune suppression to restore antitumor immune responses. The drug has been investigated in clinical trials for various brain tumors (including glioblastoma multiforme), melanoma, solid tumors, central nervous system neoplasms, ependymoma, diffuse intrinsic pontine glioma (DIPG), medulloblastoma, atypical teratoid rhabdoid tumor (ATRT), brain metastases, recurrent malignant gliomas/melanomas[1][2][3][4][5][8].
