Zamtocabtagene autoleucel is an investigational autologous chimeric antigen receptor (CAR) T cell therapy developed for the treatment of relapsed or refractory diffuse large B-cell lymphoma (DLBCL) and other B-cell malignancies. It is manufactured by modifying a patient’s own T cells to express a tandem CAR that targets both CD19 and CD20 antigens on the surface of malignant B-cells. This dual targeting aims to overcome resistance mechanisms seen with single-antigen-directed therapies by reducing the risk of antigen escape and subsequent relapse. The product is administered as a fresh (non-cryopreserved) infusion using an automated closed manufacturing system (CliniMACS Prodigy). Zamtocabtagene autoleucel has shown high response rates in clinical trials for patients who have failed at least two prior lines of therapy[1][4][5][8].
